Tetracyclines are broad-spectrum bacteriostatic antibiotics that inhibit protein synthesis by binding to the 30S ribosomal subunit and blocking aminoacyl-tRNA attachment. Agents such as tetracycline, doxycycline, and minocycline demonstrate activity against Gram-positive and Gram-negative bacteria, atypical pathogens, protozoa, and certain spirochetes. They are widely used in respiratory infections, acne therapy, zoonotic infections, sexually transmitted diseases, and malaria prophylaxis. Tetracyclines possess anti-inflammatory properties, enhancing their usefulness in dermatology and chronic inflammatory disorders. Resistance through efflux pumps, ribosomal protection proteins, and enzymatic inactivation remains a challenge. Newer derivatives like tigecycline expand activity against resistant organisms, maintaining the clinical relevance of this antibiotic class.
