Antibiotic anticancer drugs are APIs derived from microbial metabolites that exert cytotoxic effects through DNA intercalation, free radical generation, or inhibition of topoisomerase enzymes. This class includes anthracyclines (doxorubicin, daunorubicin), bleomycin, mitomycin C, and actinomycins. They are among the most potent chemotherapeutic agents, widely used in leukemia, lymphoma, breast cancer, sarcomas, and various solid tumors. Their mechanisms disrupt DNA integrity, inhibit replication, and trigger apoptosis. However, cumulative toxicity-such as cardiotoxicity with anthracyclines and pulmonary toxicity with bleomycin-requires careful dosing and monitoring. Despite toxicity concerns, antibiotic anticancer drugs remain central to modern chemotherapy regimens.
