β-Lactamase Inhibitors

β-Lactamase Inhibitors

β-Lactamase inhibitors are APIs designed to block bacterial β-lactamase enzymes that degrade β-lactam antibiotics, thereby restoring or enhancing antibiotic activity. These agents, including clavulanic acid, tazobactam, sulbactam, and novel non-β-lactam inhibitors, work by forming stable complexes with β-lactamases or irreversibly inactivating their catalytic sites. Their use significantly expands the spectrum of penicillins and cephalosporins against resistant pathogens producing TEM, SHV, CTX-M, or carbapenemase families. β-Lactamase inhibitors are essential in combination products to treat respiratory infections, urinary tract infections, intra-abdominal infections, and hospital-acquired resistant organisms. With rising antimicrobial resistance, next-generation inhibitors targeting metallo-β-lactamases and serine carbapenemases continue to be developed.
