Nitrofurans are synthetic antimicrobial agents characterized by a nitrofuran ring that undergoes intracellular reduction to generate reactive intermediates capable of damaging bacterial DNA, ribosomal proteins, and metabolic enzymes. These drugs, including nitrofurantoin, furazolidone, and nitrofurazone, exhibit broad-spectrum activity against Gram-positive and Gram-negative bacteria, particularly in urinary and gastrointestinal infections. Nitrofurantoin remains a first-line therapy for uncomplicated urinary tract infections due to its high urinary concentrations and limited systemic distribution. Nitrofurans act through multiple mechanisms, reducing the risk of resistance, although long-term use may be limited by pulmonary, hepatic, or neurological toxicity. Despite safety considerations, nitrofurans remain valuable in both human and veterinary medicine due to their efficacy and low cross-resistance potential.
